Hydrafinil (also known as 9-fluorenol, 9-hydroxyfluorene, fluorenol) It is an analogue of modafinil that acts as a weak inhibitor of dopamine reuptake [1].

Hydrafinil potentially promotes wakefulness without altering the subject's sleep schedule

Although pharmacokinetic data for hydrafinil remain limited, the same anecdotal testimony suggests that the wake-enhancing effects of the investigated nootropic do not come at the cost of interfering with the subject's sleep schedule [4]:

“T+4.2h – The dosage loses effectiveness and there is a return to the initial value. There is no accident of any kind to write home about, but simply a return to basic vigil…” [sic]

This may be explained by the more potent but shorter duration of Hydrafinil. Subjects report that Hydrafinil causes a greater increase in cognition than Modafinil, but that it lasts only 4 to 6 hours, compared to Modafinil's 12+. Hydrafinil's shorter half-life means it is potentially much less likely to interfere with the sleep schedule of a human subject.

Hydrafinil can potentially be used for polyphasic sleep cycles

Anecdotes from research subjects on Reddit promote hydrafinil as a way to enter polyphasic sleep: the practice of sleeping for multiple periods throughout an entire day. Some biohackers use the polyphasic sleep pattern to complete more tasks. Hydrafinil has given at least one subject the boost in wakefulness they needed to adapt to this way of resting [5]:

“One person stayed awake for 40 hours taking hydrafinil taking it every four hours. At 35 hours, the person was still functioning normally and had Hydrafinil. At 36 hours, the effect of the hydrafinil wore off and the person became extremely sleepy and tired and began hallucinating due to extreme lack of sleep. Then the person took hydrafinil and it worked fine for the last four hours. The person even helped someone with homework and played video games as if he were rested. The person crashed 40 hours after the hydrafinil wore off and was sleeping for many hours.” [sic]

While the polyphasic sleep pattern may not be for everyone, this may indicate another possible research application of Hydrafinil.

Hydrafinil is potentially even less addictive and more effective than modafinil in promoting alertness

Like Modafinil, Hydrafinil is not a stimulant in the traditional sense. It is a weak dopamine transporter (DAT) agonist, meaning it can increase alertness with minimal risk of addiction.6]:

“Fluurenol belongs to a class of drugs known as eugeroics. These are stimulants that produce long-lasting mental excitement that leads to wakefulness. The idea behind the production of these drugs is to improve alertness without necessarily causing peripheral bodily effects or, better yet, abuse/tolerance/addiction of common stimulants.

And Fluorenol is no different. The stimulation of dopamine pathways by hydrafinil is much less substantial than that of modafinil, meaning it is less likely to be addictive.”

Pharmacology of hydrafinil

Researchers have speculated that Hydrafinil's primary mechanism of action is actually very different from Modafinil, Adrafinil, and related analogues.11]:

“Dunn [8,9] reported that fluorenol, which is the 9-hydroxyfluorene derivative formed from 9-fluorenyl-S(=O)-CH2-C(=O)-(4N-piperazine-1N)- C(=O) -CH3 itself shows a stronger eugeroic effect but a 59% weaker DAT binding capacity than modafinil. This result is unusual as fluorenol is very different structurally from modafinil, indicating a mechanism quite different from that of modafinil.

Since the fluorene moiety can readily accommodate electron binding, an investigation of electron binding to fluorenol showed no evidence of elongation of the fluorene C9-OH bond, ruling out the formation of a fluorene radical anion species similar to shown by 9-fluorenyl-S. (=O)-CH2-C(=O)-(4N-piperazine-1N)-C(=O)-CH3. However, an examination of electron binding to the protonated fluorenol species 9-fluoreneOH2+ showed an elongation of the fluorene-OH2+ bond, indicating a dissociative electron-binding reaction.” [sic]

Hydrafinil's main action does not come from binding to DAT and inhibiting dopamine uptake, so it is thought to be less addictive than other DAT agonists.

The dissociative electron bonding reaction is a way to absorb free radicals. The buildup of free radicals in the brain during the day is thought to be one of the many drivers of sleep.12]:

“This study demonstrates that examination of modafinil and modafinil-like analogs that have been shown in the literature to exhibit cognitive and wake-promoting psychobiological properties produce free radicals when subjected to electron binding. "Such observations are consistent with a cognitive impairment and eugeroic mechanism of free radical oxidative stress for modafinil and its related analogues."

In other words, the metabolism of modafinil and its analogues shows that, in addition to DAT binding, the presence of free radicals in the brain drives sleep and removal of these free radicals can reverse this action.

“A meta-analysis of published studies on sleep deprivation and oxidative stress in the brain was used to test the hypothesis that sleep is a dynamic resting state with antioxidant properties. Wakefulness is thought to involve high neuronal metabolism and potentials.” neuronal electrical, and requires high levels of oxygen and, therefore, oxidants.

Sleep is a state with increased antioxidant activity that promotes protection of the brain against free radicals by reducing the production of oxidants. “ROS (reactive oxygen species) and other markers of oxidative stress can accumulate in the brain during wakefulness and therefore behave as sleep promoters.”

These data, together with the observation of a dissociative binding mechanism in Hydrafinil, suggest that the eugeroic effect of Hydrafinil comes from its ability to absorb free radicals in the brain. This action is important as Hydrafinil is reported to be a more potent eugeroic without the side effects of many other eugeroics available as prescription medications or research chemicals.

Applying this data to practical use, many subjects report that Hydrafinil takes between 30 and 60 minutes before taking effect and lasts a total of 6 to 8 hours. For this reason, Hydrafinil is likely to have a shorter half-life compared to Half-life of modafinil 12-15 hours.
